              <p>Alzheimer's disease (AD) is the most common fonn of dementi a in UlC eldcrly affccting more thun 35 million indil'iduals worldwidt'. AD is associated with memory and cognitive decline. Apolipoproteins are known to be implicated in AD. Among them, the apo1ipoproteins E (ApoE) and J (ApoJ) arc known to be tightly associated to the pathophysiology of AD. Both ApoE and ApoJ have nemoprotective and antioxidant activity. Apolipoprotein D (ApoD) could also display antioxidant activity and its presence may rcducc the leve! of' lipid peroxidation. While apolipoprotein D (ApoD) expression is highcr in AD brains, its role in AD and particularly in spatial memory and in leaming remains to be investigated. Our hypothesis is that ApoD plays an important role in neuroprotection and that its increase would be a compensatory mcchanism. Our goal is study the ent':ct of ApoD in the spatial mcmory and learning. To do so, intracerebroventricular injections (icv) of streptozotoc.in (STZ) were pcrfom1cd in 2 groups of micc: wild-typc (wt) and knock-out ApoD (ApoD-KO) at 8 months ok!. Icv-STZ model is known to induce neuropathological and spatial memmy alterations in mice, oxidative stress and tau hyperphosphorylation. Memory and leaming ofmice were evaluated by performing two behavioral tests: Y-Maze and Morris \Vatcr Maze (MWJVI) 3 weeks aftcr injection. Detlcits of spatial mem01y were observed in ApoD-KO mice following an icv-STZ as compared ApoD-KO with saline injection. Western-blot analysis showcd an increase of ApoJ in the striatum from wt mice icv-STZ and from ApoD-KO mice. Other markers rci:Hcd to oxidativc stress, ncurodegeneration and inflammation are perf(mned. In conclusion, ApoD could play a role in spatial memory. We highlight a compensatory regulation between bath antioxidant ApoD and ApoJ. Support: NSERC. Chaire Louise and Ardré Charron on Alzheimer's Disease.</p>
